The White Sox announced the moves before Sunday's 6-3 loss to Minnesota.
Manager Ozzie Guillen says he expects Castro "to be out for a little while because that's his throwing hand."
Minnesota Twins second baseman Alexi Casilla was batting when Castro missed a Mark Buehrle pitch with his glove and it hit his right hand. He was immediately replaced by A.J. Pierzynski. Castro was batting .235 with four home runs and 10 RBIs in 23 games.
Guillen says Flowers will play a similar role backing up Pierzynski.
Flowers, who was the starting catcher with the Charlotte Knights, has to adjust to only getting occasional starts as Pierzynski's back up.
"I'll just keep up my routine like I'm playing every day, doing the same early work," Flowers said. "Hopefully that will be enough to carry over."
Flowers was hitting .261 with a .390 on-base percentage and 15 home runs and 32 RBIs in Triple-A.
